Well, not really. I mean, not without some sophisticated diagnostic tools. You would have to have a tool that will allow you to command gear changes and stuff like that. Also, you would need a pressure tester. Now you might be able to get your hands on one of these. This is not a compression tester, but a pressure tester that is used to check the line pressure of the ATF running throughout the tranny.
The tranny can be removed from the car without dropping the subframe. Of course, you will have to pull the engine first. Several round here have done it.
And ya know, I was thinking about your problem the other day and got to thinking. I was saying that it might be your pump, right? Well, what I failed to mention is that your pump is actually driven, pressure wise, by the torque convertor. If there is something mechanially wrong with your TC, that would lead your pump into a state of not pumping.

Still hard to tell though.
