As soon as you remove the valve core, all your existing refrigerant will escape.

Take necessary precautions to avoid freeze-dried eyeballs, fingers, or anything else you plan on keeping by evacuating the system first.

Once opened and repaired, the system should be put under a hard vacuum, then recharged with refrigerant to spec.
