Air leakage in a sealed enclosure isn't a big problem if the box is built right in the first place. Some caulking (liquid nails, silicone, etc...) along the joints would be the best bet. Also make sure you pre-drill holes for the screws the sub is mounted with. If the holes are not predrilled you may get some air leakage around the sub, since the surface will warp as the screw goes in.

I usually lay a small round foam gasket around the inside edge of the sub to prevent leakage as well.

IMHO its not worth the money to buy dynomat just to use inside a sub enclosure. But then again, its your money.
